ABSTRACT

To investigate the protective effect of Yiqi Huoxue decoction on the kidney in diabetic goto-Kaizaki (GK) rats. Eight Wistar rats were used as normal control group. According to body weight and blood glucose, 16 GK rats were randomly divided into model control group and Yiqi Huoxue group, with 8 rats in each group. Drugs were administrated for 6 weeks. The rats were weighed and fasting blood glucose (FBG) were measured weekly; the glycosylated hemoglobin (HbALc) and 24 h metabolic rate were measured at the end of third and sixth weeks, at the same time, urine was collected to measure urinary glucose (U-GLU), urinary creatinine (U-CREA) and total proteinuria (U-TP) by biochemical method and calculate 24 h protein excretion rate (UAER). At the end of treatment, rats were sacrificed in anesthesia, and creatinin (CREA), uric acid (UA) and cholesterol (CHOL) were detected by biochemical method and glutathione peroxidase (GSH-Px) was detected by manual method with the blood drawn from heart. Both kidneys were taken and weighed to calculate the renal hypertrophy index. The kidneys were placed into neutral formalin and observed under light microscope for pathological change after Periodic Acid-Schiff (PAS) staining. The results showed that as compared with the normal control group, FBG, HbALc, 24 h Metabolic rate, U-GLU, UAER, serum CREA, UA and CHOL levels, as well as renal hypertrophy index were significantly increased (<0.05) in the model group, while the body weight growth rate, serum GSH-Px level were significantly reduced(<0.05); in addition, renal glomerular injury and structural changes were observed in the model group. As compared with the model group, serum GSH-Px level was significantly increased (<0.05) in the Yiqi Huoxue group, While FBG, HbALc, 24 h Metabolic rate, U-GLU, UAER, serum CREA, UA and CHOL levels, as well as renal hypertrophy index were significantly reduced (<0.05); meanwhile, renal glomerular injury and structural changes were improved in the Yiqi Huoxue group. The results showed that Yiqi Huoxue decoction can significantly increase serum GSH-Px level, reduce FBG, HbALc, 24 h metabolic rate, U-GLU, UAER, serum CREA, UA and CHOL levels, as well as renal hypertrophy index, and improve renal pathology in diabetic GK rats.

ABSTRACT

BACKGROUNDOral squamous cell carcinoma (OSCC) is one of the most common malignancies in the oral and maxillofacial region. Yes-associated protein 1 (YAP1) has been implicated as a bona fide oncogene in solid tumors. We seek to elucidate the role of YAP1 in OSCC tissue.

METHODSWe identified YAP1 gene and protein overexpression in 30 OSCC patients and 10 normal oral mucosa tissues by immunohistochemistry, Western blotting and reverse transcription polymerase chain reaction (RT-PCR).

RESULTSIn the normal oral mucosa by immunohistochemical staining, YAP1 mainly located in both the cytoplasm and nucleus mainly the nuclei of the basal cells. In OSCC, the expression of YAP1 translocated from the nucleus to cytoplasm; YAP1 being mainly located in both the cytoplasm and nucleus of the adjacent mucosa. The expression of YAP1 gradual increased in normal oral mucosa, tumor adjacent mucosa and low grade, middle grade, high grade OSCC tissue by Western blotting. Significant difference was found between the expressions of the normal oral mucosa and OSCC tissue (P < 0.05). The coincidence was detected between the normal oral mucosa and OSCC tissue by RT-PCR (P < 0.05).

CONCLUSIONSYAP1 is involved in the carcinogenesis and development of OSCC. There is a transformation between nucleus and cytoplasm.

ABSTRACT

OBJECTIVETo investigate antagonism effects of total flavonoids from Chrysanthemum morifolium. (TFCM) against lead induced oxidative injury.

METHODNinety male mice were randomly divided into 9 groups. Mice except normal control group inject lead acetate every other day for 20 days. In the next 10 d, drugs were orally administrated to mice once a day. After the last aministration, mice were sacrificed and immediately subjected to necropsy. The concentration of lead, zinc and copper in blood, brain, liver and kidney were determined. The body weight, relative organ weight, antioxidant enzyme levels (GSH, GSH-Px, SOD and CAT) and lipid peroxidation products (MDA) were performed.

RESULTTFCM might antagonize the decrease of body weight and the increase of organ weight/body weight ratio. The combined treatment with TFCM and DMSA can significantly lower the lead levels in blood, brain, liver and kidney. In contrast, lead concentration in mice treated with TFCM alone did not show significant change in these organs. The other trace elements such as zinc and copper had no significant decrease after TFCM or DMSA treatment. Middle and high-dose TFCM was more effective than DMSA in increasing the activity of GSH, GSH-Px, SOD, CAT and decreasing the concentration of MDA in mice brain. In addition, high-dose TFCM was more effective than DMSA in increasing the activity of GSH-Px, CAT and decreasing the concentration of MDA in mice liver and kidney. The combined treatment with TFCM and DMSA also can reverse lipid peroxidation and increase antioxidant enzyme levels in lead poisoning mice dose-dependently, and it had more beneficial effects than treatment with DMSA alone.

CONCLUSIONTFCM might improve antioxidant defense system, reverse lipid peroxidation and protect brain, liver and kidney against lead induced oxidative damage in mice significantly.

ABSTRACT

OBJECTIVETo study the effects and its mechanisms of arsenic trioxide (As2O3) on cell growth and human telomerase reverse transcriptase (hTERT) of human tongue cancer cells (Tca8113 cell line).

METHODSThe growth inhibition rates of Tca8113 by various concentrations of As2O3 were detected by MTF method. Cell apoptosis was detected by FCM labeled with Annexin V-FITC. hTERT gene expression was detected by RT-PCR method. hTERT protein of Tca8113 cells was determined by Western blot assay.

RESULTSThe results showed that As2O3 could inhibit the growth of Tca8113 effectively and apoptotic rate of Tca8113 cells was obviously increased in a dose and time-dependent manner. (83.40 +/- 7.31)% cells treated with 5 micromol/L As2O3 were inhibited on 72 hour point, the early apoptosis rate reached on 26.40% +/- 3.42% at that time. Moreover hTERT mRNA and protein were decreased depended on the dose and time of As2OC3, mRNA expressions of hTERT in test groups were greatly lower than that of control group on 72 hour point.

CONCLUSIONIt was suggested that As2O3 could significantly inhibit the growth of Tca8113 cells by inducing causing cell apoptosis and down-regulating the expression of hTERT mRNA gene and protein which might be one of its action mechanisms.

ABSTRACT

OBJECTIVE: To study the abnormality of p53 and H-ras gene in laryngeal cancer cells and their relation to the infection of human papillomavirus(HPV) subtypes. METHODS: The HPV subtypes, P53 and H-ras gene were examined in 28 cases of laryngeal cancer by polymerase chain reaction (PCR), single strand conformation polymorphism(SSCP), restriction fragment length polymorphism (RFLP) and DNA sequencing. RESULTS: P53 gene mutation existed in 50%cases of laryngeal cancer and DNA sequencing showed base substitution, insertion or deletion in 4 cases; The positive rate of the H-ras oncogene mutation was only 3.57% All high risk HPV infections (HPV-16 or-18) were distinctly seen in the cancer cases, especially in cases with P53 mutation. CONCLUSION: The high risk HPV subtype infection and P53 gene abnormality may play an important role in laryngeal cancer progression and these two factors may be correlated to each other.

ABSTRACT

OBJECTIVE: To assese the value and clinical significance of bioactive material-hydroxyapatite (HA) combined with TGF-beta1 in bone defect healing. METHODS: The applicability of bioactive material-HA granules combined with TGF-beta1 (group A) as a substitute for bone graft was observed in SD rat model. The results were compared with those of HA granules (Group B) and ungrafted bone defect (Group C). RESULTS: Group A demonstrated the highest level of type I collagen mRNA during healing period. CONCLUSION: The bioactive material-HA combined with TGF-beta1 can efficiently bond to ongrowing new bone comparing to HA granules.
